摘要 PROBLEM TO BE SOLVED: To provide a method for precisely determining the base sequence of DNA, by transcribing a nucleic acid in the presence of RNA polymerase, a DNA contg. a promoter sequence and an inorganic pyrophosphatase, and reading out the produced nucleic acid. SOLUTION: This method for determination of the base sequence of DNA comprises reacting ribonucleoside 5-triphosphate including ATP, GTP, CTP and UTP and derivatives thereof with one or more kinds of 3- deoxyribonucleotide-5-triphosphate including 3d ATP, 3d GTP, 3d CTP, 3d UTP and derivatives thereof in the presence of RNA polymerase and the DNA fragment contg. a promoter sequence for the RNA polymerase to yield product of nucleic acid transcription, separating the product of nucleic acid transcription, and finally, reading out the sequence of the nucleic acid from the fraction obtained. Thus, the method enables precise determination of the base sequence of DNA through the nucleic acid transcription in the presence of an pyrophophatase.
