PROBLEM TO BE SOLVED: To simply and properly discriminate the genotype of virus by synthesizing a DNA complementary to the virus DNA from the DNA extracted from the infected tissue by using a specific oligonucleotide as a primer, and analyzing the synthesized DNA by an electrophoresis. SOLUTION: The genotype of a virus, especially cucumber mosaic virus(CMV) is simply and properly distinguished by synthesizing double stranded DNA having a complementary sequence and a homologous sequence to the base sequence of a virus RNA from the RNA extracted from a virus infected tissue by using an oligonucleotide having a sequence selected from among sequence groups of formulas I to III as a primer, and analyzing the synthesized DNA by an electrophoresis to visually detect the kind of the virus. The distinguishing method is useful for effective prevention of the CMV infecting many field crops and causing mosaic disease, and efficient development, etc., of a resistant crop. |
