A nucleic acid construct which includes a nucleotide sequence encoding a site-specific RNA polymerase, a promoter including a recognition site for said site-specific RNA polymerase, the promoter being upstream relative to said nucleotide sequence but in reverse orientation, and a cloning site which, relative to said nucleotide sequence, is upstream of the promoter. Nucleic acid may be cloned into the cloning site for assessment of activity under test conditions of a promoter within the insert. Host cells and methods allow for identification and isolation of promoters active under test conditions, monitoring of promoter activity, and screening for substances which modulate promoter activity.
