PROCESS FOR PREPARING A HIGHLY PURIFIED STANDARDISED CONCENTRATE OF HUMAN VON WILLEBRAND FACTOR WITH HIGH PERCENTAGE OF HIGH-MOLECULAR MULTIMERS AND A CONCENTRATE PREPARED THEREBY

A process for preparing a highly purified standardised concentrate of human von Willebrand factor with high percentage of high-molecular multimers from a plasma cryoprecipitate fraction. The plasma is purified by aluminium hydroxide. The process comprises a combination of three chromatographic separation steps whereas the first two steps represent a ion exchange chromatography treated with vinyl-type polymeric resin containing pores of a high diameter, i.e. pores with molecular weight of 10exp(6) daltons, with diethylaminoethyl groups; and the third step represents an affinity chromatography on gelatine- -Sepharose gel. The concentrate prepared thereby exhibits a high specific activity over 300 U CBA/mg of the protein or 180 U RCo/mg of the protein. The concentrate's activity-to-antigen ration is at least 1.5 and the content of multimers of high molecular weight is at least 65 percent.
