PROBLEM TO BE SOLVED: To obtain a new DNA encoding glucuronic acid transferase polypeptide having a specific amino acid sequence and useful for producing an enzyme for synthesizing HNK-1 epitope specifically expressed in nerve cells and immunocytes, etc. SOLUTION: This DNA having a base sequence encoding glucuronic acid transferase polypeptide having an amino acid sequence of the formula has the following characteristics: having an action on transferring glucuronic acid from a glucuronic donor to a glucuronic acid receptor; specifically transferring glucuronic acid to the N-acetyllactosamine residues of asialo orosomucoid and nerve cell adhesive molecule; having an optimal reaction pH of about 6.0-6.5 (100 mM MES buffer solution, 37 deg.C); being activated with Mn<2+> , wherein the activity is maintained in the coexistence of 5 mM neolactotetraose-phenyl-C14 H29 ; and having a mol.wt. of about 45,000 (SDS-polyacrylamide electrophoresis) or about 90,000 (gel filtration). The DNA is useful for producing an enzyme for synthesizing NHK-1 epitope related to autoimmune diseases, etc. |
