PROBLEM TO BE SOLVED: To obtain an optimum sample introduction time and a sample introduction interval with one preliminary test and to determine the minimum number of columns accurately by introducing a small amount of sample into a column and measuring the absorbance of an eluate. SOLUTION: An elution curve is created by measuring the absorbance of an elution liquid by introducing a small amount of sample into a column. A peak timeμn ' and a dispersionσn ' of each constituent to be separated are obtained from each dilution curve. Then, based on the dilution curve of an expression, a sample introduction time h and a sample introduction time interval T from the start of sample introduction and to a next start of sample introduction are changed for simulation, thus obtaining the maximum sample introduction time and the minimum sample introduction interval where a constituent to be separated can maintain an allowable purity. In the expression, m indicates a sample introduction count to one column, a indicates a constitution number to be separated,μn is equal toμn '+h/2, andσn <2> is equal toσn'<2> +h<2> /12. This method eliminates the need for performing a preliminary test for optimizing separation conditions many times.
