METHODS AND COMPOSITIONS FOR TARGETED DNA DIFFERENTIAL DISPLAY

Methods and compositions are described for distinguishing between the expression of genes in two or more biological samples. In one embodiment, a PCR-based genomic differential display method is described. The method uses PCR suppression to reduce genome complexity and to amplify genomic fragments containing a targeted interspersed repeat sequence. The PCR products can be displayed by size to create a genomic fingerprint or cloned to create a library of fragments containing the targeted repeat sequence and their flanking sequences. The primers and method of the present invention allows for the accurate identification of differentially expressed gene(s) in various cell types.
