PROBLEM TO BE SOLVED: To count web-like red blood cell, red blood cell and platelet in a whole blood sample quickly with high sensitivity by determining the total number of the web-like red blood cell, red blood cell and platelet using a fully automated analyzer based on the measurement of scatter and absorption of light. SOLUTION: In order to count and characterize cells preferably within 20-30 sec of initial reaction on the aliquot of a whole blood sample, a web-like red blood cell staining reagent containing a cationic dye is employed at a concentration effective for staining the web-like red blood cell in the whole blood sample. At first, aliquot in a blood sample is conditioned along with a cationic dye of about 6-20μg/ml, an amphoteric surfaceative agent of sphering agent of about 4-148μg/ml, and an aqueous reagent component containing a solution which contains a buffer for sustaining pH of the composition at about 7.2-7.8 to prepare a reaction mixture. The reaction mixture is then passed, cell by cell, through an optically irradiating region and the light being scattered and absorbed during an interval between two angles are detected for each cell thus identifying web-like red blood cell, red blood cell and platelet and to obtain the total number of cells. |
