PROBLEM TO BE SOLVED: To simply and accurately detect the mutation of a gene by reacting a target gene for detecting its mutation with a gene probe, subjecting the reaction product to a reaction for taking a labeled substrate in the presence of a polymerase. SOLUTION: This method for detecting the mutation of a gene comprises hybridizing a target gene for detecting its mutation, such as a gene containing human K-ras gene codon 12, with a gene probe set so as to contain a base corresponding to the mutated site of the target gene at the 3'-terminal or its near site in the sequence, such as a gene probe having a bare corresponding to the point mutation site of the human K-ras gene codon 12 at the 3'-terminal or its near site, subjecting the hybridization product to a reaction for taking a labeled substrate in the presence of a DNA polymersase, and subsequently analyzing the reaction for taking the labeled substrate. Thereby, the mutation of the target gene such as a genome gene, a cancer gene, a viral gene or a bacterial gene can simply and accurately be detected. |
