PROBLEM TO BE SOLVED: To obtain the subject new gene encoding trehalose phosphorylase containing a specific amino acid sequence as a partial amino acid sequence and useful for producing trehalose capable of sweetening drinks and foods without causing their discoloration and denaturation. SOLUTION: This new gene encodes trehalose phosphorylase containing amino acid sequences of formula I and formula II or amino acid sequences homologous to the amino acid sequences of formula I and formula II as partial amino acid sequences. The gene is used for producing the trehalose phosphorylase useful for the production of the trehalose capable of sweetening drinks and foods without causing their coloration and denaturation. The enzyme is obtained by extracting the whole DNA from Grifola frondosa CM-236 (FERM 35), subjecting the extracted whole DNA to synthesis and amplification treatments in the presence of a DNA polymerase by the use of the partial gene of the trehalose phosphorylase, inserting the obtained gene into a vector, and subsequently expressing the gene in a host cell.
