PROBLEM TO BE SOLVED: To secrete and produce a different kind of protein by culturing Aspergillus oryzae host transformed with a vector containing DNAs which are used for the stimulation of expression, the secretion of the expressed product and the protein and can be inserted into the genome of the Aspergillus oryzae. SOLUTION: This method for expressing and secreting a protein product with Aspergillus oryzae comprises transforming the Aspergillus oryzae not having a functional gene for a selected marker with a recombination DNA- cloning vector and subsequently culturing the transformed host in a culture medium. The recombination DNA cloning vector contains a DNA sequence whose one or more copies can be inserted into the genome of the Aspergillus oryzae host and which encodes a function for stimulating the expression of the gene, a marker suitable for selecting the transformed cells, a preliminary sequence for providing the secretion of the expressed product into the culture medium, and a DNA sequence encoding the desired protein product. |
