Preparation of anti-Rh globulin. When injected intramuscularly into a woman with a different blood group from her husband, 72 hr. after birth of first child, the anti Rh globulin stops the formation of antibodies in the blood of the mother and hence prevents the occurrence of erythroblastosis foetalis in a second child during pregnancy. The anti-Rh globulin is obtained from human plasma by fractionating with a suitable solvent. The solvent of choice is methanol, and the fractional precipitation is carried out at as low a temperature as possible in order to avoid denaturation of the protein, but at the same time, since the solubility of the plasma components is also dependant on temperature, a temperature must be chosen which will allow the undesired components of the plasma to go into solution. |
