This invention relates to an isolated DNA comprising a sequence that encodes a uridine diphospho-N-acetylglucosamine:polypeptide beta -N-acetylglucosaminyl transferase (O-linked GlcNAc transferase, OGT), as well as vectors comprising the DNA suitable for the expression of OGT, and host cells harboring the vector that express OGT. The invention further relates to an isolated protein exhibiting O-linked GlcNAc transferase activity and having the amino acid sequence of an O-linked GlcNAc transferase. The invention additionally relates to a method of expressing a protein exhibiting O-linked GlcNAc transferase activity and having the amino acid sequence of an O-linked GlcNAc transferase. Furthermore the invention relates to a method of identifying an inhibitor of O-linked GlcNAc transferase. Additionally the invention relates to methods of assessing the status or predisposition of a patient to pathologies associated with OGT activity. |
