摘要 PROBLEM TO BE SOLVED: To provide a new gene coding an enzyme protein that has an amino acid structure corresponding to a glucose dehydrogenase in which a specific amino acid residue is substituted with another amino acid and needs pyrroloquinolinequinone as a coenzyme originating from Escherichia coil, and useful as an enzyme for determination of glucose. SOLUTION: This is a new gene coding a modified glucose dehydrogenase(GDH) that uses pyrroloquinolinequinone(PQQ) as a coenzyme or a novel plasmid including the this gene where in the PQQGDH, the 775 amino acid, namely histidine residue in the amino acid sequence from 772-778, -Ala-Gly-Gly-His-Gly- Ser-Phe- is substituted with an amino acid selected from among asparagine, aspartic acid, serine, tyrosine or lysine and is useful in the production of an enzyme for determining glucose in clinical tests and food product analyses. This gene is prepared by site-specific mutation of the structural gene of PQQGDH originating from Escherichia coli at a specific site.
