PROBLEM TO BE SOLVED: To obtain the subject DNA useful for producing a heparitinase useful as a research reagent for in vivo functions of heparan sulfate or a heparin or for analysis of the substance in a biological component. SOLUTION: This DNA is capable of coding a polypeptide of a heparitinase having at least a part of an amino acid sequence represented by the formula. The DNA is obtained by directly extracting the whole DNA from a microbial cell of a Bacillus circulans HpT298 strain according to a conventional method, transferring the extracted DNA into a suitable vector, transferring the resultant transfer vector into a host, preparing a gene library for the whole DNA of the Bacillus circulans HpT298 strain, estimating the base sequence from a partial amino acid sequence of a heparitinase protein, preparing a synthetic nucleotide probe, selecting a clone containing the DNA of this invention with the prepared synthetic nucleotide probe and isolating the objective DNA from the obtained clone. |
