A high performance liquid chromatography method. The method employs an HPLC apparatus composed of a series of chromatography segments connected end-to-end by valved connectors which are operable to communicate adjacent segments, or to divert fluid from a joined segment to an elution collection site. In the method of the invention, a sample is applied to the segments, in an amount sufficient to saturate stationary-phase material in upstream and intermediate segments with bound sample compounds, with the most hydrophobic compounds saturating the stationary phase material in the most upstream segments. Elution of compounds from each segment individually yields at least one eluted sample with the desired compound in purified form.
