Novel 2-aralkoxy-benzamide derivatives of the general formula wherein A is a mono- or di-alkylamino radical, a radical of the formula in which p and q are each 1 or 2 and Q is a sulphur or oxygen atom or a methylene, -NH or -NR- group, in which R is alkyl or a radical of the formula in which m is 1 or 2 and R is a C 1-5 alkyl group; W, Y and Z are each a hydrogen or halogen atom or an amino or C 1-5 alkylamino radical; X is a hydrogen or halogen atom; and n and k are each 1 or 2; and acid addition and quaternary ammonium salts thereof are prepared by reaction of a 2-aralkoxy-benzoic acid derivative of the general formula wherein B is a chlorine atom or a C 1-5 alkoxy group, with an amine of the general formula H 2 N-(CH 2 ) n -A, followed optionally by conversion of the product to an acid addition or quaternary ammonium salt thereof. 2 - Benzyloxy - 3,5 - dichlorobenzoyl chloride is prepared by treatment of 2-benzyloxy-3,5- dichlorobenzoic acid with thionyl chloride. Pharmaceutical compositions having local anaesthetic properties comprise, as active ingredient, a 2-aralkoxy-benzamide derivative of the first general formula above or a pharmacologically acceptable acid addition or quaternary ammonium salt thereof, together with a pharmaceutically acceptable carrier.</p> |
