PROBLEM TO BE SOLVED: To obtain the new DNA comprising a gene having a specific base sequence and coding human alanine transferase, capable of being released in serum on hepatic diseases such as viral hepatitis or cirrhosis, and useful for producing the enzyme for diagnosing the serums of the hepatic diseases, etc. SOLUTION: This new gene DNA has a base sequence of the formula, codes the amino acid sequence of human ALT (alanine aminotransferase), and is useful for efficiently producing the human ALT for diagnosing the serums of the hepatic diseases. The DNA is obtained by using a plasmid containing a gene coding the cloned human ALT as a template, amplifying plural gene fragments by a PCR using plural primers in order to substitute five amino acid cordons in the gene of the human ALT, and subsequently substituting the five regions of the human ALT gene with the plural gene fragments. |
