<p>A modified human alanine aminotransferase (ALT) gene wherein codons of five amino acids in a human ALT, i.e., from the methionine (Met) of the initiation codon, the 4th, 5th, 7th, 39th and 222nd amino acid residues have been substituted respectively by codons of methionine (Met), serine (Ser), threonine (Thr), aspartic acid (Asp), glycine (Gly) and alanine (Ala) and, at the same time, restriction enzyme sites have been added to the upstream and dowstream of the gene. An activated human ALT having the same properties as those of the authentic enzyme can be efficiently produced by culturing E. coli which has been transformed by a recombinant plasmid constructed by ligating the above-mentioned modified human ALT gene to a vector.</p> |
