PROBLEM TO BE SOLVED: To obtain chitinases having an excellent stability at a high temperature and suitable for a large scale production of chitooligosaccharides from chitin. SOLUTION: The heat resistant chitinases II, III and IV have the following enzymatic properties. (1) Action: to hydrolyze chitin to form chitooligosaccharides. (2) To hydrolyze substrate-specific colloidal chitin to form N-acetylglucosamine and N-acetylchitobiose, to hydrolyze glycol chitin to form reducing sugars and to hydrolyze p-nitrophenyl-di-N-acetyl-β-D- chitobiocide to form p-nitrophenol. (3) Optimum pH: about 10.0. (4) Optimum temperature: about 80 deg.C. (5) Molecular weight (SDS-polyacrylamide gel electrophoresis): chitinase II, about 76,000; chitinase III, about 66,000; chitinase IV, about 59,000. |
