Cloning vectors for positive selection of transformants

Cloning vectors permitting single or dual positive selection (PS) of transformants comprise two identical plasmid replicons arranged as inverted repeats to form a palindromic DNA sequence, which is interrupted at both ends of the inverted repeats by unique DNA sequences. In single-PS vectors, one of the unique DNA sequences comprises a selectable marker and the other comprises a selectable marker between two symmetrical polylinkers constituting a positive selection multiple cloning site (PSMCS). In dual-PS vectors, each inverted repeat includes an identical selectable marker and the two unique DNA sequences each comprise a different selectable marker between two symmetrical polylinkers and thus constitute two different PSMCSs.
