<p>A method of assay of the physical condition of a sample of biological cells in vitro, especially for distinguishing between cells which are proliferating, apoptotic or undergoing primary necrosis, and especially for use in samples in microtitre plates, the method involving a bioluminescent reaction of the luciferin/luciferase/cellular ATP type, wherein advantage is taken of the drop in initial light emission, representing ATP concentration, to measure ADP, by adding reagent(s) which convert ADP to ATP and measuring the emitted light intensity (or its change with respect to time) before and after the conversion of the ADP to ATP, and thereafter using information such as the ADP:ATP ratio obtained from emitted light intensity measurements to estimate cell condition. A kit comprising luciferin, luciferase ADP-converting reagent(s) and buffer required to enable assays to be carried out at volumes of not more than 240 νl is useful for the assay.</p> |
