PROBLEM TO BE SOLVED: To obtain the new subject enzyme of producing polysaccharide sulfates, consisting of a polypeptide of keratan sulfate 6-sulfotransferase and having sulfate group-transfer action from a sulfate group donor to the galactose residue of keratan sulfate. SOLUTION: This new enzyme is the polypeptide of a keratan sulfate 6- sulfotransferase having action capable of transferring the sulfate group from a sulfate donor to the galactose residue of keratan sulfate and does not transfer the sulfate group to chondroitin, chondroitin sulfate A, chondroitin sulfate C, dermatan sulfate and completely desulfated, N-sulfated heparin (CDSNS-heparin), and 6.2-6.5 optimum pH, increased in its activity by Mn<2+> or Ca<2+> , having about 2×10<-7> M (3'-phosphoadenosine 5'-phosphosulfate) Km and containing at least a part of the amino acid sequence expressed by the formula. The enzyme is used for functional analyses, productions, etc., of polysaccharide sulfates such as keratan sulfate and useful as a medicinal agent or a diagnostic agent, etc., for human diseases caused by lower degrees of sulfation, etc., of the galactose residue at 6 position in keratan sulfate. |
