<p>This invention provides a method for specifically inhibiting the activity of an enzyme for which there exists a known polypeptide inhibitor comprising a polypeptide having the sequence a1, a2...an and a suitable reactive moiety operably affixed to the alpha-carbon atom of the carboxy-terminal an residue, which method comprises contacting the enzyme with a polypeptide inhibitor comprising a polypeptide having the sequence an, an-1...a1 and a suitable reactive moiety operably affixed to the alpha-amino group of the amino-terminal an residue, wherein 10 ≥ n ≥ 2. This invention also provides a pharmaceutical composition for treating a subject inflicted with a disorder treatable by administering a known polypeptide enzyme inhibitor. This invention further provides a method of treating an inflicted subject with the instant pharmaceutical composition. Finally, this invention provides N-terminally modified polypeptide enzyme inhibitors.</p> |
