PROBLEM TO BE SOLVED: To obtain a chimera antibody molecule which contains a chimera antibody molecule having the specificity to TNF-α, and is useful in treatment of septicemia, shock, cachexia, allergy, autoimmune diseases and the liek with low antigenicity in human and reduced side effects. SOLUTION: This is a novel chimera antibody molecule having specificity to human tumor necrosis factor α (TNF-α) where the antigen-bonding site originates from a Rodent animal and the remaining immunoglobulin molecule, from human. This antibody molecule is weakened its antigenicity to human and useful for treatment of septicemia, septicemic shock, endotoxin shock, cathexia, AIDS, allergy, tubercolosis, blood coagulation disorders and autoimmune diseases. This chimera antibody molecule is obtained by incorporating the cDNA coding the heavy and light chains in the variable parts in a murine anti-human TNF-α antibody and a cDNA coding the heavy and light chains in the stationary parts of human immunoglobulin into a vector and allowing the vector to express in the host cells. |
