摘要 <p>1531498 Cyanogen halide derivatives of #-1,3- glucans; carrier ligand complexes TAKEDA YAKUHIN KOGYO KK 25 Nov 1975 [26 Nov 1974 (2) 21 Oct 1975] 48349/75 Headings C3U and C3H [Also in Division C2] Water-insoluble #-1,3-glucan materials, activated for the attachment of functional, e.g. biologically active substances are obtained by reacting a water-insoluble #-1,3-glucan with a cyanogen halide, preferably in water wherein alkali is gradually added e.g. at a rate such that the pH increases at the rate of from 0À2 to 0À5 pH unit/minute to achieve a final pH of 11-13. The glucan may be of microbiological origin, e.g. as produced by micro-organisms of the genus Alcaligenes or Agrobacterium or it may be the polysaccharide pachyman which occurs in the crude drug Poria cocos. The glucan may be in powder form or in the form of a shaped article, e.g. as fibre, film, bead, ball or microcapsule. Water-insoluble carrier-ligand products suitable e.g. for affinity chromatography can be obtained by reacting the activated #-1,3-glucan with substances containing primary or secondary amino groups, e.g. enzymes, proteins, peptides, amino acids, enzyme substrates or inhibitors, antigens, antibodies or hormones. In the examples, activated glucans are coupled with p-aminophenethyl alcohol, L-leucylglycyl glycine, serumγ-globulin and insulin and with the following enzymes: pronase,α-amylase,α- chymotrypsin, wheat germ acid phosphatase, jack bean urease, and withα-amino acid ester hydrolases obtained by elaboration of the microorganisms-Acetobacter pasteurianus (IFO 3223, ATCC 6033), Acetobacter turbidans (IFO 3225, ATCC 9325), Mycoplana sp. (IFO 13213, ATCC 21759), Gluconobacter suboxidans (IFO 3432, ATCC 621) and Xanthomonas sp. (IFO 13215, ATCC 21764).</p>
