PURPOSE:To make it possible to accurately measure the concn. of an antigen or antibody, in immunological analysis utilizing red corpuscle hemolytic reaction, by adding an immobilized red corpuscle or a red corpuscle ghost membrane to serum to be examined to inhibit hemolysis caused by a non-specific hemolytic factor. CONSTITUTION:Either one of or both of an immobilized red corpuscle obtained by treating the red corpuscle of an animal species pref. same to the red corpuscle used with formalin or glutaraldehyde in hemolytic reaction or a red corpuscle ghost membrane obtained by rupturing a red corpuscle through the treatment due to saponin or acetic acid while applying centrifugal separation to the treated red corpuscle is used in inhibiting non-specific hemolysis. For example, the aforementioned inhibitor is added to serum to be examined to be reacted therewith and, thereafter, the reaction mixture is centrifuged at a low speed while an antibody-containing solution and a complement- containing solution are added to the obtained supernatant solution and, after reaction, the absorbancy of hemoglobin in the supernatant solution is measured at a wavelength of 416mn. The measured value is collated with a calibration curve obtained by similarly measuring an antigen with a known concn. to acculately know the concn. of the antigen in a specimen. |
