Method for determining presence or localisation of one or more genes or specific DNA sequences (A), or molecules (I) reactive with DNA, on a DNA (B), comprises: (i) fixing and 'combing' a quantity of (B) on a combing surface; (ii) reacting (B) with probes (II), linked to (I) or (A), and (iii) collecting information about the position of (II), distance between (II) and/or amount of (II) (for quantifying the number of hybridised (II)) so as to deduce the presence, localisation and/or quantity of genes or (A). Also claimed are: (1) a kit for carrying out the process above; (2) a genomic DNA, or fragment, able to react, under the conditions of molecular combing, with a (II) corresponding to a translation, transcription or regulatory product, and (3) molecules (III) reactive with genomic DNA that can be combed.
