Separation of mixtures of optical isomers on a simulated mobile bed consisting of an assembly of columns (C1-C8) arranged in series, containing a chiral stationary phase and which are distributed in at least three zones (I-III). In the entrance to the first zone (I), an eluant is injected and at the exit flow is withdrawn consisting predominantly of the most retained isomer. The mixture to be separated is injected into the second zone (II). The exit from the third zone consists mainly of the least retained isomer. The flows are arranged so that, compared to a reference state in which: (a) the most retained optical isomer is completely desorbed in zone (I) and completely adsorbed in zone (III) and the least retained isomer is completely desorbed in zone (II); and (b) the quantity of material loaded is at a maximum; the new process is as follows: (i) the quantity of material injected for separation is greater than that of the reference state; and (ii) if it is desirable to at least maintain the purity of the least retained enantiomer, the extract flow is greater than that of the reference state or if it is desirable to maintain the purity of the most retained enantiomer, the extract flow is below less than that of the reference state.
