PROBLEM TO BE SOLVED: To obtain a protein having a specific molecular weight, breaking activity on both N-acetyl-YVAD-methylcoumarinamide and N-acetylDEVD- methylcoumarinamide and interleukine-lβ-converting enzymelike activity. SOLUTION: This protein has ECE-like activity obtainable from a human keratinized cell, 80kDa relative molecular weight, breaking activity on both N-acetyl-YVAD-methylcoumarinamide and N-acetyl-DEVD-methylcoumarinamide and the following properties. The optimum pH: 7.5. Inhibition of protease activity: inhibited by both Ac-DEVD-CHO and Ac-YVAD-CHO, by N-ethylmaleimide and iodoacetic acid as a thiol alkylating agent and shows clear resistance up to 5mM concentration. |
