PREPARATION OF HYBRIDOMA PRODUCING ANTIBODY BINDING TO ICAM-L OR ITS FUNCTIONAL FRAGMENT

PROBLEM TO BE SOLVED: To obtain the subject cells which can produce an antibody for the ICAM assay by immunizing specific cells expressing human ICAM-1 to an animal, fusing its spleen cells to myeloma cells and selecting the desired antibody-producing strain. SOLUTION: After an animal is immunized with the cells expressing the functional fragment of human ICAM-1 which is soluble under the physiological conditions, contains amino acid sequences represented by the formula which defects the trans membrane domain, contains amino acid sequence represented by the formula of extracellular domains 1-5 and holds the ability of a vector of the immunological reactivity and the ICAM-1-dependent adhesion properties. Then, the spleen cells of the animal are fused to myeloma cells to form hybridoma cells producing the antibody, the hybridoma cells are screened to select the cell strain which can produce an antibody which can bond to ICAM-1 whereby the objective hybridoma cells are obtained.
