NEW STRAIN AND PRODUCTION OF THEOBROMINE USING THE STRAIN

PURPOSE:To profitably obtain the subject compound useful as a diuretic and an intermediate for medicines by culturing the variant of a Pseudomonas microorganism having an ability to produce theobromine from caffeine and subsequently collecting the product. CONSTITUTION:A microorganism [e.g. Pseudomonas.putida IF-3 (FERM P-12172)] belonging to the genus Pseudomonas having an ability to produce theobromine from caffeine is cultured in a medium until the logarithmic multiplication period, collected centrifugally, and suspended in a 50mM tris- maleate buffer solution (pH 6.0). The suspension is mixed with N-methyl-N'- nitro-N-nitrosoguanidine as a mutagen and subsequently allowed to stand at 30 deg.C for 30min. The microorganisms are collected and cultured in an agar medium to obtain a variant lost an ability to decompose the theobromine. The variant is cultured in a medium containing caffeine at 30 deg.C for 8hr to highly efficiently obtain the objective theobromine.
