PROBLEM TO BE SOLVED: To obtain the subject medicinal agent non-neurotoxic to hosts, capable of ceasing the protein synthesis in neuro cells through infection on the neuroma cell, also sensitive to acyclovir, by using a specific vector afforded by herpes simplex virus. SOLUTION: This apoptosis agent contains a vector obtained by removing γ1 34.5 gene from the gene of herpes simplex virus. The vector is e.g. R3616 as the recombinant virus of herpes simplex virus (HSV-1-d-34.5) which is pref. obtained by removing the 1kb base pairs in the DNA sequence containing the 34.5 gene in HSV-1 (specifically, it is recommended that the point corresponding to the 28th amino acid site in the γ1 34.5 gene is cut by the use of restriction enzyme BstE II along with cutting the 3'-terminal of the gene by the use of restriction enzyme Stu I. The dose of the vector is pref. 10 or more per oncocyte. This agent can be manufactured into a preparation by formulating the above vector with a stabilizer such an human serum albumin. |
