PCAPS VECTOR WHICH IS POSITIVE SELECTIVE VECTOR ON CAPS GENE AND USE THEREOF

PROBLEM TO BE SOLVED: To obtain a new vector containing a modified cap gene in which the expression thereof is fatal to an adenylate cyclase-positive host cell in a host containing a vector having a suitable cleavage site in a selective gene in which the functional expression thereof is interfered by insertion of an exogenote. SOLUTION: This new vector comprises a modified cap gene as a selective gene, having positive selection possibility for cloning a DNA sequence and capable of coding a CAP protein having a modified DNA-binding specificity in which the expression thereof is fatal to an adenylate cyclase-positive host cell in a host cell transformed with a vector having a suitable restriction cleavage site in a selective gene in which the functional expression thereof is interfered by the insertion of an exogenote. The vector has a possibility of the positive selection for cloning the DNA sequence in the host cell transformed with the vector and capable of cloning the DNA sequence by replicating the vector in the host cell.
