PROBLEM TO BE SOLVED: To provide an improved method for reverse transcription by which cDNA is effectively prepared from mRNA, and the obtained cDNA has a complete length because the mRNA is allowed to perform the reverse transcription over the whole length even if a long chain mRNA is used as a replica by using the reverse transcriptase at a temperature not forming the mRNA into the secondary structure. SOLUTION: A cDNA is prepared from an mRNA by performing reverse transcription of the mRNA at a temperature of 45-90 deg.C at which the mRNA does not form a secondary structure, by using a not-thermostable reverse transcriptase in the presence of one or more kinds of material having chaperone activities selected from saccharides such as trehalose, an amino acid such as N<e> -acetyl-β-lysine, a polyfunctional alcohol (derivative) and a chaperone protein such as the chaperone protein of thermoduric bacteria.
