Method for sealing and isolating an internal space (3) of a hydrodynamic rotating machine (1) having an envelope (2) delimiting the internal space (3). The machine has a shaft (6) mounted in the envelope (2) and passing through at least a wall (2a) of it. Interposed between the shaft (6) and the wall (2a) is a seal having at least a joint (8) having an annular surface (8a) opposite the shaft (6) and co-axial to it. The method is characterized in that: - formed around the shaft (6) in an adjacent position with respect to at least a part of the joint (8), and co-axial with the shaft (6) and to the surface (8a), is a throat (19,19') in the wall (2a) of the envelope (2). This throat is linked to a pressurized fluid source (25) via a valve (24); - inside the throat (19,19'), and around the shaft (8) is a toroidal flexible sealed joint (20) which can be radially deformed; - the throat (19.19') is fed with pressurized fluid at the external periphery of the joint seal (20) so as to compress the joint (20) radially so as to put it in sealed contact with the rotating shaft (8).
