PROBLEM TO BE SOLVED: To induce the nicking with a restriction endonuclease and amplify a target nucleic acid sequence according to a strand displacement amplification(SDA) by transducing an α-boronated deoxynucleoside triphosphate into a double- stranded recognition site with a restriction enzyme. SOLUTION: This method is to amplify a target nucleic acid sequence according to an SDA and comprises transducing an α-boronated deoxynucleotide triphosphate (e.g. 2'-deoxycytidine α-boronated triphosphate) into a double- stranded recognition site of a restriction endonuclease, thereby producing a semimodified restriction endonuclease recognition site capable of transducing a nick during the SDA, thereby inducing the nicking of a double-stranded DNA with the restriction endonuclease and amplifying the target nucleic acid sequence according to the SDA. |
