<p>An immortalized human hepatic cell line is disclosed. The cell line is created by lipofection of primary cultures of normal human fetal hepatic cells with a plasmid containing the second and third exons of the human c-myc gene driven by an SV-40 promoter. Cytochemical, morphological and phenotypical analyses indicate that the resulting cell line has hepatocyte characteristics. When these cells were analyzed after 90 passages they continued to express selected hepatocyte markers, including albumin and gamma-fibrinogen. Transmission electron microscopy reveals that the cells exhibit epithelial features, including desmosomal junctions and numerous surface microvilli, and form structures that resemble bile canaliculi. Methods of making the disclosed immortal hepatic cell line are also disclosed.</p> |
