Determining very low concentrations of nucleic acid by amplification

Determination of nucleic acid (I), present at low concentration in a test sample, involves amplification reaction with (a) unlabelled amplification primer (P1) and (b) labelled detection primer (P2), plus preferably polymerase and/or ligase. P2 hybridises to the amplification product (AP) and the decrease in concentration of P2 and/or the increase in concentration of AP is determined as a measure of the presence of (I).
