SOLUTION: The extent of possibility forming a substantially complementary double between a region of base sequence of mRNA and other region different from the above region in each region of a base sequence of mRNA is converted to a numerical value based on a fact that the extent of possibility forming the double chain is maximum when a distance between substantially complementary base sequences holds 3-10 bases or the extent of the possibility is converted into a numerical value based on the fact that a binding energy in forming the double chain becomes a numerical value calculated based on the closest base pair model or the extent of the possibility is numerically converted based on above both facts and a nucleic acid having a base sequence substantially complementary to a region low in the numerical value is prepared as an antisense nucleic acid to efficiently provide the objective antisense nucleic acid used for antisense nucleic acid method useful for preparation, etc., of a medicine or a gene recombinant plant.
