<p>PROBLEM TO BE SOLVED: To obtain embryonic stem cells which are prepared from an inbred strain mouse and are useful for the preparation of a gene knock-out mouse or transgenic animal. SOLUTION: The objective embryonic stem cells are prepared from an inbred strain mouse selected from C3H/HeN, C57BL/6N, DBA/1J or BALB/c and cell C-2 (FERM BP-5933), C-6 (FERM BP-5934), B6-26 (FERM BP-5935), DB1 (FERM BP-5931), Cr/A-3 (FERM BP-5932). A couple of a male and a female mouse in these strains are allowed to naturally mate and the female mouse is uterectomized on gestation day 3. The placental bulla from the uterus is cultured, then transplanted to the feeder cells after hatching and cultured to peel off inner cell aggregate. The aggregate is made to smaller cell aggregate in a small drop of a trypsin solution of about 0.25% concentration and this operation is repeated about 10 times to establish the objective embryonic stem cell.</p> |
