PROBLEM TO BE SOLVED: To measure glucose concentration in a whole blood sample by detecting the intensity of the light having a specified wavelength reflected from at least a part of a matrix. SOLUTION: The reflective index is automatically read at prescribed time intervals by a suitable device such as diffuse reflection photometer, and the change is calculated to output glucose concentration in a blood sample. Such a device reads the reflectance of the test surface of a strip in about 635nm and about 700nm. A matrix effectively separate erythrocyte and hemoglobin from the whole blood sample, and the separated membrane captures erythrocyte. The reduction in reflectance in 700nm is performed by a chromophore, but the reduction in reflectance in 700nm observed from a test surface after 15 second from reflectance switching requires the simulation of the color of the blood. The magnitude of reduction in reflectance in 635nm in a suitable time after starting the timing order becomes the scale of glucose concentration in the whole blood sample.
