The invention refers to a recombinant substrate for systems to assay in vitro agrecanase, containing a group of structural elements comprising CD5 signal sequence, FLAG epitope, for monoclonal antibodies M1 detection, human agrecane inter-globular domain, human IgG1 hinge region, human IgG1 CH3 region;; a DNA coding recombinant substrate; host vectors and cells containing such DNA and recombinant substrate use to study agrecanase activity, in order to detect new enzymatic cleavage sites, to purify agrecanase by chromatography, in order to clone agrecanase cDNA by functional cloning, to explore for an agrecanase inhibitor; a method to watch an osteoarthritis progression starting, and a diagnostic auxiliary means containing recombinant substrate.
