摘要 PROBLEM TO BE SOLVED: To easily extract with a high sensitivity a DNA useful for e.g. environmental cleanup diagnosis from microorganisms in soil, by washing soil with a weakly acidic aqueous solution containing a compound such as inorganic or organic salt or urea followed by washing the resultant soil with an aqueous solution of powdered milk. SOLUTION: A DNA in microorganisms in soil is directly extracted for the purpose of easily assaying in high sensitivity such microorganisms useful for environmental cleanup, through the following consecutive processes: 1st washing process: soil is washed with a weakly acidic aqueous solution containing an inorganic salt such as sodium nitrate or ammonium chloride, a organic salt such as ammonium acetate, potassium acetate or sodium acetate, or a compound such as urea in a concentration of 0.05-5M; 2nd washing process: the resultant soil is further washed with an aqueous solution of powdered milk such as skim milk or powdered milk for nursery use; and, if needed, optionally, bacteriolysis process to lyze microorganisms in the resultant soil by using e.g. sodium lauryl sulfate, protein-denaturing and removing process using e.g. phenol, and/or precipitation process for the objective DNA by using e.g. ethanol.
