PROBLEM TO BE SOLVED: To provide a determination method, for humanα-fetoproteins(AFT's), in which an agglutination reaction due to a nonspecific reaction is eliminated, in which a precise measurement can be performed even in a low-concentration region, whose sensitivity is high and which is quick. SOLUTION: In a determination method for AFP's, an insoluble carrier in which an anti-AFP antibody is sensitized is mixed with a sample, the change amount of the absorbance of a reaction solution due to an antigen-antibody agglutination reaction is measured, the AFP's in an unknown sample are determined by using a working curve, latex particles are used as the insoluble carrier, the insoluble carrier in which the anti-AFP antibody is sensitized is used in such a way that its concentration in an agglutination reaction becomes at 0.01 to 0.08wt.%, polyethylene glycol in an average molecular weight of 3000 to 1000000 and/or poly(vinylpyrrolidine) in an average molecular weight of 300000 to 1000000 are added in such a way that their concentration in an agglutination reaction solution becomes at 0.1 to 5wt.%, the insoluble carrier and the sample are reacted for five seconds to 15 minutes and at a constant temperature, and an increase in the absorbance due to the progress of a latex agglutination reaction is measured.
