摘要 PROBLEM TO BE SOLVED: To analyze a nucleic acid through such processes that a doubldstranded DNA specimen is treated with plural restriction enzymes and the difference between both the 3' terminals of the resultant DNA fragments is identified using plural primers, and these fragments are selectively separated by complementary strand synthesis followed by measuring the lengths of the fragments, and the distribution pattern of the lengths is detected. SOLUTION: A double-stranded DNA specimen 40 is cut with plural restriction enzymes into double-stranded DNA fragments 41-1 to 41-3, the difference between the base sequences at both the 3' terminals of these fragments is identified by the respective base sequences of the 1 to 4 bases at the respective 3' terminals of at least two kinds of DNA primers 49', 49", and the DNA fragments each with the base sequences at both the 3' terminals specified are subjected to complementary strand synthesis using the above two kinds of DNA primers in the respective vessels 55-2, 55-3 and selectively separated, and the reaction products are separated by electrophoresis and the respective lengths of the products are measured to detect the pattern of distribution of the above DNA fragment lengths to enable the long double-stranded DNA specimen to be tested, thus accomplishing the objective nucleic acid analysis.
