PROBLEM TO BE SOLVED: To obtain an antibody which is bonded selectively to Glu<17> -osteocalcin and which is not bonded to the Glu<17> -osteocalcin by using the peptide fragment of the Glu<17> -osteocalcin described in an arrangement number 5 as an antigen, preparing a composite substance with carrier protein if required, and inoculating an animal with the substance so as to be immunized. SOLUTION: The fragment peptide of Glu<17> -osteocalcin for an immune antigen is prepared by using a peptide synthesizer by an ordinary method, it is decoupled by using a glutaraldehyde or the like in the ratio of one to five times the amount of carrier protein such as bovine serum albumin or the like, and an immune animal such as a mouse or the like is dosed hypodermically with the composite substance normally one time each for two to five weeks. Then, blood is sampled from the immunized animal, serum is separated, an antibody is purified from the serum by a usually used method such as ammonium sulfate salting-out, ion-exchange chromatography or the like, and a polyclonal antibody is obtained. |
