PROBLEM TO BE SOLVED: To provide a method capable of controlling a temperature of a speci men at a prescribed reacting temperature while preventing evaporation of mois ture about a finer amount of specimen, by e.g. a PCR (polymerase chain reac tion) method. SOLUTION: A reaction of a reactive mixture is advanced by varying the temperature of the reactive mixture reserved in a substantially sealed reaction vessel so as to have a space in an upper part in the vessel with passing of time. In this case, a temperature 1 of the reactive mixture in the reaction vessel and a temperature 2 of at least a face opposing to the surface of the reactive mixture on an inner wall of the reaction vessel attaching to the space are controlled so as the temperature 2 to be higher than the temperature 1 and so as the temperature 1 to vary in linking with the temperature 2. The second objective incubator has a reaction vessel comprising a base plate having plural holes for receiving the reactive mixture and a pair of temperature controllers 21 and 22 comprising Peltier elements for varying the temperature of the reaction vessel. |
