PROBLEM TO BE SOLVED: To obtain a novel compound: lebrastatin which is useful as a medicine, particularly useful as a carcinostatic agent because it has an inhibitory action of the cell cycle by acting on the Rb protein to inhibit its phosphorylation. SOLUTION: This novel compound: lebrastatin is represented by the formula or its salt. This compound has the following physical and chemical properties: description: a white powdery substance; melting point: 222-231 deg.C; solubility: soluble in methanol and dimethyl sulfoxide, insoluble in n-hexane, chloroform and water; molecular formula: C29 H44 N2 O8 ; molecular weight: 548 (according to the FAB mass spectrometry). This substance is prepared by culturing a microorganism in Streptomyces capable of Levulastatin, for example, Streptomyces hygroscopicus subsp. hydroscopicus SANK 61995 (FERM BP-5140) and collecting the objective substance from the culture mixture. |
