PROCESS FOR MODIFYING THE ENZYME 7 beta -(4-CARBOXYBUTANAMIDO) CEPHALOSPORINACYLASE AND PURIFYING SAID ENZYME IN A SINGLE CHROMATOGRAPHIC STEP

Process for producing a modified enzyme 7 beta -(4-carboxibutane-amide) cephalosporinase which may be purified in a single chromatographic step. The enzyme production process comprises mutagenizing the gene which codes for the enzyme Acinetobacter sp. ATCC 53891 by inserting a sequence of nucleotides which codes for six histidine residues; fusing the mutant gene with high efficiency promoter DNA sequences; transforming with the constructed genic fusion cells of Escherichia coli; cultivating the transformed Escherichia coli cells; and collecting the enzymes through the use of media which contain metal chelates. The 7-aminocephalosporanic acid is an important intermediary for the fabrication of a wide variety of antibacterial agents of the family of cephalosporines.
